The Aurka cDNA was inserted downstream of tet-responsive ColA1 sequences using KH2 mouse embryonic stem (ES) cells following the strategy reported by Beard et al. 2006, Genesis 44, 23–28 [PubMed]. Tetracycline-responsive elements were inserted in the endogenous Aurkb gene either in the presence [Aurkb(loxtet)] or absence [Aurkb(tet)] of loxP sequences. Overexpression of Aurora B results in genomic instability and increased tumor susceptibility.

The Cdk4 R24C mutation was found in melanoma patients. Cdk4 R24C is insensitive to INK4 family of inhibitors resulting in increased susceptibility to tumor development. It also cooperates with the Cdk6 R31C mutation.

Knock-in of miR-203 and OSKM (Oct4-Sox2-Klf4-Myc). The OSKM inducible strain was kindly provided by Manuel Serrano´s lab in 2014. We crossed the OSKM strain with our BGC strain, knock in for miR-203. Both miR-203 and OSKM transgen are inducible by DOXYCYCLINE treatment. through the Rosa 26_rtTA allele. Reverse tetracycline transactivator (rtTA)/tetracycline-responsive element (tet-O)-driven transgene. To ensure reliable rtTA expression in a broad range of cell types, the rtTA transgene has been targeted into the ROSA26 locus.
